When you create a new layer in a file and save it back into a document, you see a small gray box appear in the lower-left corner of the work area. This is the new image layer, and after you save the edits, the image appears in the exact same location. Creating a new image layer The easiest way to create a new image layer is to press Alt+Backspace. This keystroke creates a new layer with no upper-left-hand corner. Figure 4-9 shows this keystroke. You can move this new layer to a different location in the work area, work on it, and save it to a new layer. You can also use the Layer from Background tool to create a new layer. (On a keyboard, you can press Shift+E.) When you click this tool, a layer appears over your image, as shown in Figure 4-9. You can adjust the size of the layer, which is the white square shown in the lower-right corner, but you can’t select anything in the background. If you move the cursor in the background, as shown in the figure, a red line appears over the background image. In the present study, we examined the effects of pyrrolidine dithiocarbamate (PDTC) on the development of experimental autoimmune thyroiditis (EAT) in mice. Mice were injected with 100 microg of porcine thyroglobulin (Tg) emulsified in complete Freund’s adjuvant to generate EAT. PDTC (100 mg/kg/day) was orally administered for a week before the onset of the disease and for seven weeks. EAT was evaluated by clinical signs and morphologic changes. Serum levels of autoantibodies against Tg and thyroglobulin (TgAb) in the mice were measured by enzyme-linked immunosorbent assay. Messenger RNA (mRNA) expression of cytokines in thyroid, peritoneal exudate cells (PECs), and spleen was examined by in situ hybridization. PDTC suppressed the development of EAT, as assessed by clinical signs and morphological changes, although it did not affect the levels of TgAb, compared with the control group. The ratio of CD4+/CD8+ lymphocytes in thyroid and spleen was not affected by PDTC treatment. Thyroid tissues from the control group showed an increase in mRNA expression of proinflammatory cytokines, such as tumor necrosis factor-alpha (TNF-alpha), interferon (IFN)-gamma, IL-1alpha, and IL-6, compared with the PDTC-treated group. mRNA expression of interleukin (IL)-10, an antiinflammatory cytokine, was also increased in the PDTC-treated group. PDTC-treated mice showed an increase in CD4+CD25+ lymphocytes in thyroid tissue, compared with the control group.
